What is Mesalazine?

It itself has similar therapeutic effects. 5-aminosalicylic acid, due to its active reactive groups of hydroxyl and carboxyl groups on the ring, can undergo various reactions. Therefore, in the dye industry, it can be used to manufacture various high-quality reactive dyes. Also used in the manufacturing of photosensitive paper.

 

5-aminosalicylic acid, also known as mesalazine powder, can be used as a chemotherapeutic drug for ulcerative colitis and has a significant inhibitory effect on inflammation of the intestinal wall; It can inhibit the synthesis of prostaglandins that cause inflammation and the formation of the inflammatory mediator leukotriene, thereby significantly inhibiting inflammation of the intestinal mucosa. The effect is better on the connective tissue of the inflamed intestinal wall. Used for ulcerative colitis, ulcerative proctitis, and Crohn's disease.

 

It is a peroxidase substrate suitable for ELISA analysis. The substrate generates a brown soluble end product, which can be read at 450nm by spectrophotometry. Adding 3NNaOH can terminate the reaction and then read at 550nm.

Production methods

Method 1:

After mixing salicylic acid with water, slowly add a mixture of 68% nitric acid and glacial acetic acid at 50 ℃ and stirring. Heat up again to 70-80 ℃ for reaction. Pour the reaction solution into water and place it at 5 ℃. Filter and recrystallize the filter cake with hot water to obtain solid 5-nitrosalicylic acid with a yield of 40%. Add iron powder to hydrochloric acid solution at 60 ℃. After heating and boiling, add 1/4 of 5-nitrosalicylic acid. After vigorous stirring, alternate the addition of other nitro compounds and iron powder in batches. Add and react at 102 ℃. Adjust to strong alkalinity with 50% sodium hydroxide while hot, and filter. Wash the filter cake with hot water, combine the filtrate and washing solution, add insurance powder, and acidifie with 40% sulfuric acid to a pH value of 4. Cool, filter, dry, dissolve it in hot water with sodium bisulfite, and boil it with activated carbon. Hot filtration, wash the filter cake with a small amount of hot water. Combine the filtrate and wash solution and place at 5 ℃. Filter and collect crystals, wash thoroughly with ice water, dry to obtain white needle shaped  crystals with a melting point of 278 ℃ (decomposition).

 

Method 2:

Salicylic acid is nitrated to obtain 5-nitrosalicylic acid, which then reacts with methylsulfonyl chloride to protect the phenolic hydroxyl group. It is then hydrogenated to reduce the nitro group to an amino group, and finally hydrolyzed to obtain mesalazine.

 

Method 3:

Aniline is diazotized to obtain diazonium salt, which is added to a solution of salicylic acid, sodium carbonate, and 40% sodium hydroxide. It is vigorously stirred at 0-6 ℃ while maintaining a pH value of 8. Filter, wash with saturated salt water, and press dry. Dissolve in a 40% sodium hydroxide solution and add insurance powder at 80-85 ℃. After the solution completely fades, remove aniline by steam distillation and filter. Adjust the filtrate to a pH value of 6-6.5 with concentrated hydrochloric acid and leave it overnight. Filter, wash with cold water, dissolve the dried crude product in water, add concentrated hydrochloric acid dropwise until dissolved, then add activated carbon and heat until boiling. Filter, cool, and adjust to a pH value of 2-3 with dilute sodium hydroxide. Filter, wash with cold water, vacuum dry at room temperature to obtain a gray white solid of mesalazine, with a melting point of 278 ℃ (decomposition) and a total yield of 45.9%.

 

Method 4:

Mix aminophenol and an appropriate amount of catalyst at CO2 pressure of 2.0-3.0 MPa and 130-190 ℃. Cool to room temperature, add water and insurance powder, and stir. Cool to 0-3 ℃, filter out insoluble substances, and wash with water. Combine the washing solution and filtrate, adjust to a pH value of 3-4 with hydrochloric acid, filter, wash with water, and obtain crude mesalazine. Add water, sodium bicarbonate, and sodium bisulfite to the crude product, heat until fully dissolved at 50-60 ℃, then add activated carbon and boil. Filter, cool, and adjust the dilute hydrochloric acid to a pH value of 3.5. Filter, wash, and dry to obtain a gray white high-quality product with a yield of 91.1% and a content of 99.2%.
