Animal specific antibiotics
Animal safety seriously affects human health, especially in recent years, various animal diseases have erupted around the world, such as avian influenza, swine flu, mad cow disease, foot-and-mouth disease, etc., posing a great threat to human health. However, the use of ordinary antibiotics can easily lead to the development of drug resistance in the pathogens in animals, high drug residues and toxicity, and have drawbacks such as teratogenic, carcinogenic, and environmental pollution. Therefore, developing an excellent antibiotic has become a major trend in the development of veterinary medicine in the world today. Valnemulin Hydrochloride is a new generation of semi synthetic animal specific antibiotics, belonging to the diterpenoid class and the same class of drugs as Tylenomycin. Abroad, it is mainly used for the prevention and control of mycoplasma diseases and Gram positive bacterial infections in pigs, cows, sheep, and poultry. It has the characteristics of broad-spectrum, strong antibacterial activity, excellent pharmacokinetic characteristics, low residue, and safety. In 1999, it was approved by the European Community for the prevention and treatment of swine dysentery caused by Brachyspira Hyodysenteriae infection and porcine endemic pneumonia caused by Mycoplasma pneumoniae infection. In January 2004, it was approved by the European Community for the prevention of porcine colitis caused by Brachyspirapillosicoli infection and the treatment of porcine proliferative bowel disease (ileitis) caused by intracellular Lawsonia infection. Vonimolin hydrochloride is a highly sensitive new type of animal specific antibiotic for the prevention and treatment of chronic respiratory diseases in chickens, porcine endemic pneumonia (also known as porcine asthma, porcine mycoplasma pneumonia), actinomycete pleuropneumonia, swine dysentery, porcine colon spirochemical disease (colitis), porcine proliferative bowel disease (ileitis), and other diseases. It has broad application prospects in veterinary clinical practice in China. It is reported that currently, a few powerful animal medicine production enterprises in China are applying for the registration of national Class II new animal drugs. On September 9, 2010, the Ministry of Agriculture announced in Announcement No. 1457 that the raw materials and premixes of vornimolin hydrochloride developed in China were approved by the Ministry of Agriculture as National Class II New Veterinary Drugs. Therefore, it has broad application prospects in veterinary clinical practice in China. The clinical application of its formulation will undoubtedly bring good news to the development of pig and chicken farming, and make greater contributions to the development of animal husbandry.
【 Appearance 】 White or light yellow crystalline powder with hygroscopicity; Soluble in water and anhydrous ethanol, almost insoluble in tert butyl methyl ether. The specific rotation is+15.5~+18.0, and the pH value is 3.0~6
Pharmacological action
Inhibiting bacterial protein synthesis by binding to the 50S subunit on the ribosome of pathogenic microorganisms, inhibiting RNA synthesis at high concentrations, and binding to the V region (23SrRNA) on the ribosome of pathogenic microorganisms can prevent accurate localization of transpeptidases, inhibit protein synthesis, and ultimately lead to the death of pathogenic microorganisms, exhibiting antibacterial activity.
Pharmacokinetics
Absorption and distribution: Internal absorption is rapid, and the peak time of blood drug concentration is 1-4 hours. There is a linear relationship between plasma concentration and dosage. The bioavailability is above 90%. Widely distributed in the body, the drug concentration is high in lung and liver tissues, often several times higher than the blood plasma concentration. Metabolism and excretion: Widely metabolized and excreted in the body, its metabolites are mainly excreted rapidly from feces through bile, and about 30% are excreted from urine. The half-life of plasma is 1.3-2.7 hours.
Pharmacodynamics
Valnemulin Hcl is a semi synthetic antibiotic; Derivatives of polysaccharides from shiitake mushrooms. Antibacterial. This product has a wide antibacterial spectrum and is effective against Gram positive bacteria, negative bacteria, anaerobic bacteria, mycoplasma, and spirochetes. It has a strong inhibitory effect on Staphylococcus aureus, Streptococcus, Mycoplasma pneumoniae, Mycoplasma synovial fluid, Actinobacillus pleuropneumoniae, short spirochetes suis, short spirochetes pilosum, and intracellular Lawson's bacteria, but is less effective against Gram negative bacteria such as Escherichia coli and Salmonella. The mechanism of action is to bind to the 50s subunit of ribosomes in bacteria and other microorganisms, inhibiting protein synthesis in bacteria and other microorganisms, and also inhibiting RNA synthesis at high concentrations.
Indications
Mainly used for the prevention and treatment of chronic respiratory diseases in chickens, porcine endemic pneumonia (also known as porcine asthma, porcine mycoplasma pneumonia), actinomycete pleuropneumonia, porcine dysentery, porcine colon spirochete disease (colitis), and porcine proliferative bowel disease (ileitis).
Drug interactions
1. This product can affect the metabolism of plasma carrier antibiotics such as monensin and salinomycin. When used in combination, adverse reactions such as slow growth, dyskinesia, and paralysis may occur.
2. When used in combination with antibiotics that can bind to the 50S subunit of bacterial ribosomes, it has an antagonistic effect.
3. When mixed with chloramphenicol at a ratio of 7.5:40 or doxycycline at a ratio of 7.5:25, it has a synergistic effect on the infection of respiratory pathogens in pigs.
Adverse reactions
1. The adverse reactions of pigs mainly occur in Landrace pigs from Denmark and Sweden, characterized by fever, loss of appetite, ataxia, lying down, edema or erythema, eyelid edema, with a mortality rate of less than 1%. However, if there is a secondary infection, the mortality rate can be increased.
If the feed concentration is greater than 200ppm, there will be a decrease in feed intake or unwillingness to feed in the first few days.
3. When used in combination with plasma carrier antibiotics such as monensin and salinomycin, the toxicity of the latter can be increased.
Synthetic route
Using refined truncated pleuropterin (2) as raw material, it was sulfonated with p-toluenesulfonyl chloride and reacted with dimethylcysteamine hydrochloride to obtain truncated pleuropterin cysteamine substitute; In addition, (R) -2- (1-methoxycarbonyl-2-allyl) amino-3-methylbutyrate potassium was prepared by reacting D-valine, methyl acetoacetate, and potassium hydroxide. After being activated by ethyl chloroformate, it reacted with a truncated cysteine substituted compound of pleurotus. Finally, by adjusting the pH value, reverse phase extraction was performed, and freeze-dried to obtain vornimolin hydrochloride. The specific reaction process is as follows:
Shelf life
it is sensitive to light and humidity, so it needs to be kept away from light and sealed for storage. Store in a dark and sealed environment at a temperature not exceeding 30 ℃, with a shelf life of 5 years; Made into veterinary medicine with a shelf life of 3 years; Mix in powder and store away from light and moisture, with a shelf life of 3 months; Made into granular materials, stored away from light and moisture, with a shelf life of 3 weeks.
Purpose
The mechanism of action of Wonimulin is to inhibit bacterial protein synthesis at the ribosomal level, and it also inhibits RNA synthesis at high concentrations. Mainly for antibacterial purposes, but also for sterilization at high concentrations. The antibacterial spectrum is broad, effective against G+and G - bacteria, highly effective against the genera Mycoplasma and Spirochetes, but less effective against gut bacteria such as Escherichia coli and Salmonella.
