Synthetic process
The synthetic process of Tacrolimus powder mainly includes the following steps:
Isoprene is used as a raw material to synthesize a tetracyclic intermediate through sulfuric acid-catalyzed addition reaction.
The tetracyclic intermediate is subjected to carboxylation reaction to obtain FK506 acid.
FK506 acid is reacted with isohexanoyl chloride in the presence of potassium carbonate to generate FK506 precursor.
The FK506 precursor undergoes alkylation reaction to produce Tacrolimus.
It should be noted that multiple fine separations, purifications, and structural characterizations are required in the synthesis process, and the control of process parameters is also crucial. Currently, there are many literature reports on the synthesis methods of it.
