Production process

a.Obtain sheep seminal vesicles.

b.Homogenize the vesicles in a solution containing KCl and EDTA-Na2 at pH 8 to obtain a mixture of enzymes.

c. Incubate the enzyme mixture with hydrogen peroxide, glutathione, and dihomo-γ-linolenic acid.

d. Extract the reaction mixture with acetone, ether, and dichloromethane.

e. Obtain crude prostaglandins.

f. Separate prostaglandin E1 from the crude product using a silica gel column.

• Preparation of crude PGE product using sheep seminal vesicles as raw material involves enzymatic preparation, incubation, organic solvent extraction, and silica gel column separation.

• Sheep seminal vesicles [KCl, EDTA-Na2, pH 8] → Enzyme mixture [hydroquinone, glutathione] → Reaction mixture [arachidonic acid, O2] → Solvent mixture [acetone, ether, dichloromethane] → Crude PGS product [separation] → [silica gel column] Crude PGE product.

• For the preparation of PGE1 final product, for every 1g of crude PGE product, use 20g of silica gel. Activate the silica gel by mixing with activated silver nitrate in a ratio of V ethyl acetate: V glacial acetic acid: V petroleum ether: V water = 220:22.5:125:5 (petroleum ether boiling point 90-120°C) as the developing agent. Perform column chromatography using the wet method. Dissolve a small amount of the crude product in the same developing agent, load it onto the column, and elute. Collect separate fractions of PGE1 and PGE2. Concentrate the PGE1 fraction under nitrogen at temperatures below 35°C to remove the acetic acid odor. Dissolve it in ethyl acetate, wash with acidic water at pH 4-5, and remove silver using physiological saline solution. Leave the ethyl acetate solution in the refrigerator overnight to obtain the PGE1 final product.
